Human CAPN1/calpain 1 ELISA Kit

The format is a double-antibody sandwich ELISA with colorimetric detection, delivering a sensitivity of 0.188 ng/mL and a measuring range of 0.313–20 ng/mL.

Calpain-1 is a key enzyme involved in cell signaling, cytoskeletal dynamics, and protein degradation. The manufacturer notes that dysregulation of calpain-1 activity has been linked to muscular dystrophy, neurodegenerative disorders, and cardiovascular diseases, making it a biomarker relevant to studies of those conditions and potential therapies.

Reported linearity across dilutions is 82–98% for serum, 83–99% for EDTA plasma and 80–97% for Heparin plasma. Recovery ranges are 97–104% for serum (average 100%), 91–105% for EDTA plasma (average 96%) and 91–102% for Heparin plasma (average 93%). The protocol includes reagent equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin (SABC) incubation for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after stop solution.
