Calpain 1 (CAPN1) is a calcium-dependent, non-lysosomal cysteine protease that functions as a heterodimer, with the large catalytic subunit encoded by the CAPN1 gene on chromosome 11q13. This protease cleaves a broad set of substrates involved in cytoskeletal maintenance, cell-cycle progression, and receptor-mediated signal transduction, and altered CAPN1 activity has been documented in studies of muscular dystrophy, ischemic injury, cataracts, and several tumor types. The Assay Genie immunoassay is configured to quantify the large subunit of human calpain 1 in experimental specimens and is supplied as a complete reagent set for 96 determinations in a standard microplate layout.

The assay produces a colorimetric signal suitable for readout on a standard plate reader, and the product is labeled for research use only. Researchers commonly apply this kit to lysed cell preparations, tissue homogenates, and conditioned media, and the protocol insert covers how to handle individual specimen matrices such as serum or plasma when those are used. Reconstituted standards should be aliquoted to limit repeated freeze-thaw cycles, and unused plate strips and working reagents are stored refrigerated and protected from light to preserve activity over the kit's shelf life. Each package contains a pre-coated microplate, calibrated standard curve of recombinant human CAPN1, detection antibody concentrate, chromogenic substrate, wash buffer, and stop solution, providing the consumables needed to generate a standard curve and interpolate unknown concentrations from prepared specimens.
