Description

A Sandwich ELISA Kit from Assay Genie designed for the precise measurement of caldesmon levels in human samples. Caldesmon, encoded by the CALD1 gene, is a critical regulator of smooth muscle contraction and plays a crucial role in cell motility and cytoskeletal organization. Dysregulation of caldesmon has been linked to various diseases, including cancer, cardiovascular disorders, and neurological conditions, making reliable quantification valuable for research into disease progression and potential therapeutic targets.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method with a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL, and is stored at 2–8°C for 12 months. Linearity for serum (n=5) ranges 87–104%, 82–91%, and 80–98% at 1:2, 1:4, and 1:8 dilutions respectively; EDTA plasma (n=5) at 85–101%, 84–100%, and 88–99%; and heparin plasma (n=5) at 87–104%, 83–100%, and 83–100%. Recovery averages 91% for serum (range 86–99%), 92% for EDTA plasma (range 88–95%), and 97% for heparin plasma (range 91–105%).

The assay involves equilibrating reagents to room temperature, incubating standards and samples at 37°C for 90 minutes, adding a biotin-labeled detection antibody for 60 minutes, then adding HRP-Streptavidin for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes, followed by stop solution and absorbance measurement at 450 nm.
