Human Calcineurin ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for quantitative measurement of calcineurin (CaN) in human serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Calcineurin is a key enzyme involved in immune response and neuronal signaling, and its dysregulation has been linked to autoimmune diseases, neurodegenerative disorders, and cardiac hypertrophy, making this kit a useful tool for related research.

The kit uses a double-antibody sandwich ELISA format with colorimetric detection. Reported sensitivity is 0.188 ng/mL with an assay range of 0.313–20 ng/mL. The procedure involves equilibrating reagents and TMB substrate to room temperature, incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labeled detection antibody with further incubation, adding HRP-Streptavidin (SABC), developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. Recommended storage is 2–8°C for 12 months.

Linearity testing (n=5) showed serum recovery ranges of 86–98% at 1:2, 84–101% at 1:4, and 81–92% at 1:8; EDTA plasma 89–104%, 82–96%, and 86–97%; and heparin plasma 87–99%, 85–95%, and 83–98%. Spiking recovery ranges were 90–105% (serum), 92–103% (EDTA plasma), and 85–103% (heparin plasma), with average recoveries of 96%, 98%, and 97% respectively. Included components are an ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer, and technical manual.
