Description

Manufactured by Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of the C-terminal fragment of agrin (CAF) in human samples. The double-antibody assay uses colorimetric detection and is suited to research applications where accurate quantification of CAF levels is required. CAF, also known as agrin, contributes to the formation and maintenance of neuromuscular junctions and synapses, and dysregulation of CAF has been associated with neurological conditions including myasthenia gravis and Alzheimer's disease.

The kit reports a sensitivity of 28.125 pg/mL and a detection range of 46.875-3000 pg/mL. It accepts a broad panel of human sample types, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity spans 80-104% across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, while recovery averages 92-97% in the same matrices. Samples are processed with a standard sandwich workflow: equilibration of reagents, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution addition.

Components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody, HRP-streptavidin conjugate (SABC), TMB substrate, sample, antibody, and SABC dilution buffers, stop solution, 25X wash buffer, plate sealers, and a technical manual. Unopened kit components are stored at 2-8°C for 12 months, with the plate and standard additionally suitable for 12 months at -20°C after foil-bag sealing.
