Human CA12/Carbonic anhydrase 12 ELISA Kit

Assay Genie offers a sandwich ELISA kit built on a double-antibody format for the quantitative measurement of carbonic anhydrase 12 (CA12) in human samples. The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is intended for research applications investigating CA12, an enzyme involved in regulating pH and maintaining ion balance, with dysregulation linked to cancer, kidney disorders, and osteoporosis.

Reported performance includes a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. Linearity in serum (n=5) spans 86-105% at 1:2, 87-103% at 1:4 and 85-104% at 1:8; in EDTA plasma (n=5) 84-101%, 81-100% and 85-102% at the same dilutions; and in heparin plasma (n=5) 86-101%, 88-99% and 83-95%. Recovery averages are 98% for serum, 99% for EDTA plasma and 96% for heparin plasma, with ranges of 88-101%, 90-104% and 90-101% respectively.

The workflow includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution. Components supplied are a dismountable ELISA microplate, lyophilised standard, 100X biotin-labelled antibody, 100X HRP-Streptavidin conjugate (SABC), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual. All components should be stored at 2-8°C, with the plate and standard also suitable for 12 months at -20°C.
