Assay Genie's C4c complement-fragment kit is a quantitative immunoassay built around the cleaved complement component 4c, which is generated when classical-pathway activation converts C4 into C4a and C4b, and then further processes C4b into C4c and C4d. Because C4c appears in circulation only as a downstream cleavage product, its measurement reflects ongoing classical-pathway activity rather than total C4 protein levels, which is why researchers studying complement-driven inflammation, autoimmune disease flares, and complement-deficiency disorders rely on C4c as a sensitive activation marker.

Prepared for studies in human specimens, the procedure accepts cell-culture supernatant alongside plasma-type and serum-type specimens, making it appropriate for clinical cohorts, animal-model work using human cell lines, and in vitro complement-activation experiments. The workflow follows a standard ELISA format: capture antibody pre-coated on the plate, sequential incubation with standards and samples, biotinylated detection antibody, streptavidin-HRP, and a substrate step producing a signal proportional to C4c concentration. The assay is calibrated for research use only and is intended for laboratory investigation of complement biology.

Provided as a complete kit with all reagents needed to run the plate, the product supports laboratories monitoring classical-pathway turnover in conditions such as systemic lupus erythematosus, immune-complex disease, and hereditary angioedema, and complements efforts by vaccine and therapeutics developers characterizing classical-pathway engagement by candidate antibodies and complement-targeted drugs. Samples should be handled according to standard immunological-assay practice, with appropriate refrigeration of reconstituted reagents and protection of prepared working solutions from light.
