Human C3c/Complement Fragment 3c ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative detection of C3c / Complement Fragment 3c in human samples. The double-antibody sandwich format delivers a sensitivity of 0.188 ng/mL with a working range of 0.313–20 ng/mL, and the assay uses a colorimetric TMB-based read-out with absorbance measured at 450 nm.

Validated specimen types include serum, plasma (EDTA and heparin), cell culture supernatant, cell or tissue lysate, and other liquid samples. Sample-to-sample linearity has been demonstrated across 1:2, 1:4 and 1:8 dilutions: serum 87–104%, 86–103% and 89–95%; EDTA plasma 93–101%, 83–96% and 87–101%; heparin plasma 80–95%, 81–96% and 87–96% (n = 5 per matrix). Spike-recovery performance was 95–105% (average 100%) in serum, 85–98% (average 91%) in EDTA plasma, and 90–104% (average 98%) in heparin plasma.

The procedure involves a 90-minute primary incubation at 37°C, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and immediate absorbance measurement at 450 nm after addition of stop solution. Unopened kit components are stored at 2–8°C for up to 12 months. Components supplied include a pre-coated microplate, lyophilised standard, biotin-labelled detection antibody (100X), HRP-streptavidin conjugate (100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer, and a technical manual.
