Description

A Sandwich ELISA Kit from Assay Genie enables quantitative detection of human Complement C1q tumor necrosis factor-related protein 1 (C1QTNF1), a secreted protein implicated in inflammation, adipogenesis, and energy metabolism whose dysregulation has been linked to metabolic disorders, cardiovascular disease, and inflammatory conditions. The double-antibody sandwich assay uses colorimetric readout and is validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples of human origin. Reported sensitivity is 1.875 ng/mL with a working range of 3.125–200 ng/mL, and unopened reagents remain stable for 12 months at 2–8 °C. The target corresponds to UniProt entry Q9BXJ1 and is also known by the synonyms CTRP1, GIP, UNQ310, PRO353, and C1q and tumor necrosis factor related protein 1.

Manufacturer performance data includes linearity and recovery testing across serum, EDTA plasma, and heparin plasma matrices. At 1:2, 1:4, and 1:8 dilutions, linearity is reported as 92–103%, 87–100%, and 88–103% for serum (n=5); 83–97%, 88–98%, and 83–100% for EDTA plasma (n=5); and 87–99%, 82–98%, and 81–100% for heparin plasma (n=5). Spiked recovery ranges from 89–104% (mean 94%) in serum, 88–100% (mean 94%) in EDTA plasma, and 85–103% (mean 93%) in heparin plasma.

The protocol involves equilibrating reagents and TMB substrate to room temperature, an initial 90-minute incubation at 37 °C for antigen capture, a 60-minute incubation with biotin-labelled detection antibody, a 30-minute incubation with HRP-Streptavidin (SABC), colour development with TMB substrate protected from light for 10–20 minutes, and a stop reaction followed by absorbance reading at 450 nm. Sample-specific guidance is supplied for serum (clot, then centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8 °C), tissue homogenates, cell culture supernatants, and cell lysates prepared with protease inhibitors.
