The assay uses a double antibody sandwich format and is intended for a variety of research applications. c-Src and CSK are key regulators of cellular signaling pathways, playing important roles in processes such as cell growth, differentiation, and survival, with their dysregulation implicated in diseases including cancer, inflammatory disorders, and autoimmune diseases.

The kit has a reported sensitivity of 0.469 ng/mL and an assay range of 0.781-50 ng/mL, with absorbance read at 450 nm. Reported linearity at 1:2, 1:4, and 1:8 dilutions is 85-99%, 85-99%, and 85-95% for serum (n = 5); 84-97%, 82-100%, and 86-96% for EDTA plasma (n = 5); and 82-91%, 86-97%, and 86-99% for heparin plasma (n = 5). Recovery averages 96% across serum, EDTA plasma, and heparin plasma samples, with recovery ranges of 90-105%, 86-105%, and 92-100% respectively. Reagents are stored at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, followed by primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, color development with TMB substrate in the dark for 10-20 minutes, and a stop reaction with absorbance measurement at 450 nm.
