Human c-Myc ELISA Kit

From Assay Genie comes a sandwich ELISA kit for the quantitative measurement of c-Myc protein in human samples, using a double antibody format with colorimetric detection at 450 nm. It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and should be stored at 2-8°C for 12 months.

The kit reports a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. Linearity was assessed by diluting samples at 1:2, 1:4, and 1:8 across serum, EDTA plasma, and heparin plasma matrices. Recovery testing on the same matrices gave average recoveries of 93% (serum), 96% (EDTA plasma), and 91% (heparin plasma). The c-Myc protein targeted by this kit (UniProt P01106) is described as a key regulator of cell growth and proliferation whose expression has been linked to various cancers, making it a useful biomarker for cancer biology research.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples, and blanks onto the pre-coated plate, and incubating at 37°C for 90 minutes for antigen binding, followed by biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction read immediately at 450 nm. Sample preparation includes clotting and centrifugation of serum, anticoagulant collection and centrifugation of plasma, homogenisation of tissue in PBS with protease inhibitors, and standard lysis for cell lysates.
