Human Bifunctional heparan sulfate N-deacetylase/N-sulfotransferase 2 (NDST2) ELISA Kit

Assay Genie offers a Sandwich ELISA designed for the quantitative detection of human Bifunctional heparan sulfate N-deacetylase/N-sulfotransferase 2 (NDST2), also known by synonyms including Glucosaminyl N-deacetylase/N-sulfotransferase 2, N-heparan sulfate sulfotransferase 2, NDST-2, N-HSST 2 and HSST2. The assay is reactive with human samples and has been validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, detecting both natural and recombinant forms of the target.

Reported performance characteristics include a detection range of 78–5000 pg/mL and a sensitivity of 34 pg/mL. Intra-assay precision is given as 4.5% CV and inter-assay precision as 8.0% CV. Linearity has been evaluated in serum (102–112% at 1:2; 93–102% at 1:4; 82–94% at 1:8; 101–111% at 1:16), EDTA plasma (106–115%, 94–103%, 100–111%, 100–109%) and heparin plasma (98–108%, 109–119%, 109–118%, 109–121%). Spike-and-recovery performance shows an average of 93% (range 87–99%) in serum and 95% (range 89–101%) in plasma.

NDST2 is a bifunctional enzyme that catalyses both the N-deacetylation and the N-sulfation of glucosamine (GlcNAc) within the heparan sulfate glycosaminoglycan, a prerequisite step for later modifications during heparin biosynthesis, and is required for the exosomal release of SDCBP, CD63 and syndecan. The protein (UniProt P52849) is encoded by the NDST2 gene (NCBI Gene ID 8509) at chromosomal location 10q22, has a reported molecular weight of 42,093 Da, and is localised to the Golgi apparatus membrane as a monomer.
