Description

Assay Genie offers a Competitive ELISA Kit for quantifying Human Beta-defensin 119 (DEFB119) in human research samples. The assay uses a competitive ELISA format coated with antigen and is read colorimetrically. Reported performance includes a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored sealed at 2-8°C and must not be cryopreserved.

Linearity was evaluated at 1:2, 1:4, and 1:8 dilutions. For serum (n=10) values ranged from 87-105% to 85-105% to 85-102%; for EDTA plasma (n=10) 84-97%, 82-99%, and 93-99%; for heparin plasma (n=10) 81-100%, 81-93%, and 80-97%. Recovery testing yielded ranges and averages of 86-94% (90%) for serum (n=10), 93-105% (99%) for EDTA plasma (n=10), and 89-104% (95%) for heparin plasma (n=10). The procedure includes TMB substrate equilibration, sample and biotin-labeled antibody incubation at 37°C for 45 minutes, washing, color development in the dark at 37°C for 10-20 minutes, stop solution addition (blue to yellow), and absorbance reading at 450 nm. Beta-defensins are an important class of antimicrobial peptides that play a vital role in the innate immune response, and Beta-defensin 1-19 has been shown to have antimicrobial activity against a wide range of pathogens.
