Human Beta,beta-carotene 9',10'-oxygenase (BCO2) ELISA Kit

From Assay Genie, this sandwich ELISA kit enables quantitative measurement of human Beta,beta-carotene 9',10'-oxygenase (BCO2) in serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids. BCO2, also known as B-diox-II or beta-carotene dioxygenase 2, is a mitochondrial enzyme that asymmetrically cleaves beta-carotene at the 9',10' double bond to form beta-apo-10'-carotenal and beta-ionone and also oxidatively cleaves lycopene, contributing to carotenoid and retinoid metabolism.

Specific for natural and recombinant human BCO2, for research use only, the kit reports a sensitivity of 0.23 ng/mL and a detection range of 0.78–50 ng/mL. Intra-assay CV is 6.0% and inter-assay CV is 10.6%. Spiked recovery averages 113% (range 107–119%) in serum and 115% (range 109–120%) in plasma. Dilutional linearity across 1:2, 1:4, 1:8 and 1:16 spans approximately 80–113% in serum, 104–118% in EDTA plasma and 104–120% in heparin plasma.

The assay uses an antigen-coated microplate where standards, samples and blanks are added, followed by Detection Reagent A and Detection Reagent B working solutions separated by wash steps, TMB substrate development protected from light, and absorbance reading at 450 nm.
