Human BDG/beta-D-glucan ELISA Kit

Designed for the detection of beta-D-glucan (BDG) in human samples, this Sandwich ELISA from Assay Genie employs a double-antibody format with colorimetric measurement at 450 nm. The assay is reactive with human samples and provides quantitative measurement of the target in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported performance includes a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Dilutional linearity (n=5) was confirmed in serum at 86-99% (1:2), 83-101% (1:4) and 84-99% (1:8); EDTA plasma at 80-96%, 80-99% and 83-98% respectively; and heparin plasma at 84-97%, 83-99% and 86-103%. Spike-and-recovery runs from 87-103% in serum (average 93%), 90-105% in EDTA plasma (average 96%) and 89-103% in heparin plasma (average 95%).

The workflow follows a six-step sandwich protocol: equilibrate reagents and incubate standards and samples on the pre-coated plate at 37°C for 90 minutes; add biotinylated detection antibody at 37°C for 60 minutes; incubate with HRP-Streptavidin at 37°C for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; stop the reaction and read absorbance at 450 nm. The kit ships with a pre-coated microplate, lyophilised standard, biotinylated detection antibody concentrate (100×), HRP-Streptavidin concentrate (100×), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25× wash buffer, plate sealers and a technical manual. Reagents should be stored at 2-8°C for 12 months.
