Description

A Sandwich ELISA, Double Antibody kit from Assay Genie is designed for the quantitative measurement of human BD-1/beta Defensin 1 (DEFB1; UniProt P60022) across a range of liquid sample types including serum, plasma, cell culture supernatant, and cell or tissue lysate. Colorimetric detection is carried out on a pre-coated microplate using TMB substrate, with absorbance read at 450 nm following the addition of stop solution. According to the manufacturer datasheet, the kit offers a sensitivity of 18.75 pg/mL and an assay range of 31.25–2000 pg/mL.

Linearity has been demonstrated across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with recoveries reported as 89–103% (average 98%) for serum, 86–102% (average 94%) for EDTA plasma, and 85–103% (average 96%) for heparin plasma. The procedure follows a sequential incubation format: antigen binding at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, and TMB colour development in the dark for 10–20 minutes. Reagent components include lyophilised standard, 100X biotin-labelled antibody, 100X SABC, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, and 25X wash buffer.

Unopened reagents should be stored at 2–8°C for 12 months, with the standard and coated plate additionally suitable for long-term storage at –20°C. Beta-defensin 1 is an antimicrobial peptide involved in innate immune defence and has been associated with inflammatory disease, making this kit a useful tool for biomarker and immunology research. A separately listed Assay Genie PharmaGenie ELISA Kit (SBRS0008) is also available for cross-species work in human, mouse, and rat samples; that competitive-format assay has a reported sensitivity of 0.9 ng/mL and range of 0.1–1,000 ng/mL.
