The colorimetric assay is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with absorbance read at 450 nm. The reported sensitivity is 0.469 ng/mL with a working range of 0.781-50 ng/mL. Reported linearity at 1:2, 1:4 and 1:8 dilutions for serum (n = 5) is 88-96%, 87-101% and 85-103%; for EDTA plasma (n = 5) 86-101%, 85-101% and 82-101%; and for heparin plasma (n = 5) 82-92%, 81-94% and 84-95%. Reported recovery averages are 93% for serum, 92% for EDTA plasma and 92% for heparin plasma. The protocol involves equilibrating reagents and TMB substrate to room temperature, incubating standards and samples on the pre-coated plate at 37°C, binding with biotin-labelled detection antibody at 37°C, adding HRP-Streptavidin, developing with TMB substrate in the dark, and adding stop solution before reading absorbance at 450 nm. Kit reagents are intended for storage at 2-8°C, protected from direct light where indicated, for 12 months.
