Human BAK1/BCL2-antagonist/killer 1 isoform 1 ELISA Kit

A Sandwich ELISA (Double Antibody) kit from Assay Genie designed for the quantitative measurement of human BAK1 (BCL2-antagonist/killer 1 isoform 1; Uniprot Q16611) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric TMB detection format and is positioned in the cell death research area, where BAK1 is studied as a pro-apoptotic regulator linked to cancer and neurodegenerative disease contexts.

Reported analytical performance includes a sensitivity of 0.375 ng/mL and a working range of 0.625-40 ng/mL. Spike-and-recovery values are reported as: serum (n=5) 93-103% with an average of 97%; EDTA plasma (n=5) 92-105% with an average of 98%; heparin plasma (n=5) 87-100% with an average of 93%. Linearity for serum at 1:2, 1:4 and 1:8 dilutions is reported as 86-96%, 86-96% and 86-104% respectively; corresponding EDTA plasma values are 85-99%, 83-98% and 86-99%, and heparin plasma values are 88-96%, 81-97% and 82-94%. The kit is stored at 2-8°C with a stated shelf life of 12 months.

The procedure is a standard sandwich workflow: equilibrate reagents and TMB substrate to room temperature and load standards, samples and blanks onto the pre-coated plate, incubating at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop colour with TMB substrate in the dark for 10-20 minutes; add stop solution and read absorbance at 450 nm immediately.
