This quantitative immunoassay from Assay Genie measures human B7-2, also known as CD86, a co-stimulatory membrane glycoprotein expressed on professional antigen-presenting cells such as B lymphocytes, dendritic cells, and macrophages. CD86 delivers the second signal required for naive T-cell activation through engagement of CD28, while also binding CTLA-4 with higher affinity to dampen responses, making it a focal target in studies of immune regulation, autoimmunity, transplant tolerance, tumor immunology, and vaccine adjuvant development.

The kit is configured for 96 individual determinations arranged on a single strip-plate, supporting full standard curves run in duplicate alongside multiple unknown samples. Researchers apply the assay to cell lysates, tissue homogenates, and cell culture supernatants to track CD86 expression or shedding under different stimulation conditions, for example LPS, IFN-gamma, or cytokine treatments. The procedure follows a two-antibody capture protocol with an absorbance-based readout on a standard plate reader; reagents are supplied ready-to-use or as concentrated stocks as detailed in the manual.

Each package includes a pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, calibrated recombinant standard, wash buffer, and substrate. The assay is for research use only, intended for in vitro laboratory work and not for clinical diagnostic or therapeutic decision-making. Unopened components are stored refrigerated at 4 degrees C; reconstituted working solutions and aliquoted standards should be frozen at -20 degrees C and protected from repeated freeze-thaw cycles to preserve antibody binding activity through the stated shelf life.
