Beyond its role in the β-catenin destruction complex, AXIN2 (also known as Axin-2 or Axin-like protein, UniProt Q9Y2T1) is itself a transcriptional target of β-catenin/TCF, functioning as a negative feedback regulator that is widely used as a sensitive reporter of canonical Wnt pathway engagement. Assay Genie's kit is built around a 96-well microplate pre-coated with capture antibody specific for human AXIN2, and uses a biotinylated detection antibody plus Avidin-HRP conjugate with TMB substrate, with absorbance read at 450 nm ± 2 nm. Both intra-assay and inter-assay CV values are below 10%, and spike-and-recovery testing gave average recoveries of 100% in serum, 93% in EDTA plasma, and 99% in cell culture media, with linearity maintained across 1:2 to 1:16 dilutions. The protocol shows no significant cross-reactivity with related analogues. Researchers commonly apply the kit to monitor Wnt pathway activation in colorectal carcinoma cell lines, to screen small-molecule Wnt modulators in vitro, to characterize AXIN2 expression in osteoblast differentiation where it marks committed bone-forming progenitors, and to investigate the AXIN2/p53 axis in DNA damage response studies.
