The Assay Genie AVPR1A immunoassay follows a classical sandwich ELISA workflow in which a pre-coated capture antibody immobilizes the V1a receptor from serum, plasma, cell culture supernatant, or tissue lysate, and a biotinylated reporter antibody combined with HRP-Streptavidin generates a TMB-based colorimetric signal read at 450 nm in a standard microplate reader. Assay Genie reports a working standard curve spanning 62.5 to 4000 pg/ml with a minimum detectable concentration near 37.5 pg/ml, supporting quantitative comparisons between treated and control cohorts in behavioral and cardiovascular research programs.

Recommended sample volumes are 50 ul for serum or plasma and 100 ul for cell or tissue lysate; the antibody pair shows no appreciable cross-reactivity with homologous vasopressin receptors such as AVPR1B and AVPR2 or the closely related oxytocin receptor, enabling clean discrimination of V1a-specific protein abundance. Because the V1a receptor couples to Gq/G11 and signals through the phosphatidylinositol-calcium second messenger pathway, downstream changes in receptor expression measured by this kit can be correlated with phospholipase C activation, vascular smooth muscle contraction, and glycogenolysis endpoints in vitro.

Unopened kits from Assay Genie are stored at 2-8 °C and remain stable through the labeled expiry date; opened microplate strips should be sealed with desiccant and refrigerated, while lyophilized standards in their original vial and concentrated detection reagents are aliquoted and frozen at -20 °C for long-term retention. Each box contains one 96-well plate, biotinylated antibody, HRP-Streptavidin concentrate, TMB substrate, stop solution, and wash buffer sufficient for 96 individual determinations, with the full assay completing in roughly 4 hours after sample preparation.
