Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of Human Autophagy protein 5 (ATG5) in human samples. The assay is reactive with human specimens and is intended for research use only, with applications relevant to the study of autophagy, cancer, neurodegenerative disorders, and metabolic syndromes, as ATG5 is a key protein involved in maintaining cellular homeostasis by clearing damaged organelles and proteins.

According to the manufacturer specifications, the detection range is 0.312–20 ng/mL with a sensitivity of 0.12 ng/mL, using a colorimetric detection method. Intra-assay CV is reported at 6.3% and inter-assay CV at 8.6%. The assay is validated for serum, plasma, tissue homogenates, cell culture supernatants, and other biological fluids. Reported linearity for serum ranges from 82–118% across dilutions of 1:2 to 1:16; for EDTA plasma from 80–122%; and for heparin plasma from 96–115%. Average recovery is 100% for serum (range 94–106%) and 102% for plasma (range 96–108%). The kit is specific for natural and recombinant human ATG5, with listed synonyms including APG5-like, Apoptosis-specific protein, APG5L, and ASP.

ATG5 plays a role in the apoptotic process, with expression occurring downstream of caspase activity, and is crucial for IFN-gamma-induced autophagic cell death through interaction with FADD. The protein localizes to the cytoplasm and to the preautophagosomal structure membrane as a peripheral membrane protein, and forms a conjugate with ATG12 that associates with ATG16L. UniProt accession is Q9H1Y0, with a listed molecular weight of 275, and the gene is ATG5 (NCBI Gene ID 9474). The research area is noted as cancer.
