Human ATG5/Autophagy protein 5 ELISA Kit

Designed for the accurate detection of Autophagy-Related Protein 5 (ATG5) in human samples, this Sandwich ELISA from Assay Genie is a double-antibody colorimetric assay intended for research use. ATG5 is a key player in the autophagy process, which is crucial for maintaining cellular homeostasis by eliminating damaged organelles and proteins, and dysregulation of autophagy has been linked to various diseases, including cancer, neurodegenerative disorders, and metabolic conditions, making ATG5 an important biomarker for studying these diseases and developing potential therapeutic interventions.

The kit is reactive with human samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It offers a sensitivity of 0.188 ng/mL and a detection range of 0.313-20 ng/mL. Linearity data are provided for serum (92-105% / 85-105% / 85-100% at 1:2, 1:4, 1:8), EDTA plasma (87-100% / 83-96% / 82-100%), and heparin plasma (83-91% / 81-96% / 80-98%). Recovery ranges are 93-103% (average 97%) for serum, 86-104% (average 97%) for EDTA plasma, and 94-105% (average 100%) for heparin plasma. Reagents should be stored at 2-8°C for 12 months.

The assay procedure follows a six-step sandwich ELISA workflow: equilibrate reagents and TMB substrate to room temperature and load standards, samples, blanks and controls on the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; and stop the reaction and read absorbance at 450 nm immediately.
