Human ATF6/Activating Transcription Factor 6 ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the accurate quantitative detection of human ATF6 (Activating Transcription Factor 6) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric, double-antibody sandwich detection method, with reported sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. Kits are shipped and stored at 2-8°C with a stated shelf life of 12 months under those conditions.

ATF6 is a key transcription factor involved in the unfolded protein response, helping to maintain cellular homeostasis under stress conditions. Its dysregulation has been associated with cancer, diabetes and neurodegenerative disorders, making it a useful biomarker for studying disease mechanisms and potential therapeutic targets. The kit is for human reactivity. Across dilutions of 1:2, 1:4 and 1:8, reported linearity is 85–97% / 86–103% / 87–104% for serum (n=5), 85–100% / 91–93% / 82–95% for EDTA plasma (n=5), and 82–99% / 81–90% / 83–97% for heparin plasma (n=5). Spike-and-recovery performance is 92–102% (average 97%) in serum, 86–103% (average 95%) in EDTA plasma and 86–104% (average 94%) in heparin plasma.

The protocol follows a standard sandwich workflow: equilibrate reagents and TMB substrate to room temperature; load standards, samples and blank into the pre-coated plate and incubate at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin conjugate and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm immediately.
