Description

Manufactured by Assay Genie, this sandwich double-antibody ELISA is intended for the quantitative measurement of Aspartate Aminotransferase (AST) in human samples. The kit is reactive with human specimens and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection is colorimetric, and absorbance is read at 450 nm following TMB substrate development.

The assay provides a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Linearity in serum (n=5) is 88–98% at 1:2, 90–102% at 1:4, and 86–97% at 1:8; EDTA plasma (n=5) is 82–95%, 83–98%, and 84–100% across the same dilutions; heparin plasma (n=5) is 83–99%, 83–96%, and 80–93%. Recovery averages 95% in serum (range 89–103%), 95% in EDTA plasma (range 91–102%), and 92% in heparin plasma (range 85–98%). The kit should be stored at 2–8°C for up to 12 months.

The protocol involves equilibrating reagents and loading standards, samples, and controls onto the pre-coated plate, then incubating at 37°C for 90 minutes. A biotinylated detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin for 30 minutes at 37°C. TMB substrate is added and incubated in the dark for 10–20 minutes before the stop solution is applied and absorbance is measured at 450 nm. AST is an enzyme predominantly found in the liver, heart, and muscles, and elevated blood levels can indicate liver damage, heart issues, or muscle injury, making the kit a relevant tool for related research applications.
