Human ASRGL1 ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of ASRGL1 (isoaspartyl peptidase/L-asparaginase) in human biological samples. The double-antibody sandwich format uses colorimetric detection and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. ASRGL1 is an enzyme involved in the regulation of cellular metabolism and protein degradation, and has been implicated in cancer, neurodegenerative disorders, and metabolic syndromes.

The reported sensitivity is 0.469 ng/mL with an assay range of 0.781–50 ng/mL. Linearity across dilution has been demonstrated in serum (1:2 85–102%, 1:4 88–101%, 1:8 88–104%), EDTA plasma (1:2 82–99%, 1:4 87–99%, 1:8 87–101%) and heparin plasma (1:2 82–99%, 1:4 85–98%, 1:8 87–99%; n = 5 each). Recovery averages 93% for serum (range 86–105%), 97% for EDTA plasma (range 91–105%) and 97% for heparin plasma (range 90–102%).

The procedure involves equilibrating reagents and TMB substrate to room temperature, a 90-minute primary incubation at 37 °C, 60-minute biotinylated detection antibody binding, 30-minute HRP-Streptavidin (SABC) binding, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. Components supplied include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample/antibody/SABC dilution buffers, stop solution, wash buffer (25X), plate sealer and a technical manual.
