Description

A sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human Aspartoacylase (ASPA) in serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids. The assay uses a colorimetric detection method and is reactive with human samples. It is intended for research use only.

Aspartoacylase is an enzyme that catalyses the deacetylation of N-acetylaspartic acid (NAA) to produce acetate and L-aspartate. NAA occurs in high concentration in the brain, where its hydrolysis is thought to contribute to the maintenance of intact white matter; in other tissues ASPA acts as a scavenger of NAA from body fluids. Defects in ASPA are linked to Canavan disease, a rare neurodegenerative disorder, supporting the kit's positioning for neuroscience research.

Manufacturer-stated performance figures include a detection range of 78-5000 pg/mL, a sensitivity of 39.8 pg/mL, an intra-assay CV of 5.5% and an inter-assay CV of 8.1%. Specificity is reported for natural and recombinant human Aspartoacylase (UniProt P45381), described as a homodimer localised to the cytoplasm and nucleus. Recovery data are provided for serum (average 90%, range 84-96%) and plasma (average 92%, range 86-98%), and linearity is documented for serum, EDTA plasma and heparin plasma across dilutions from 1:2 to 1:16.
