The Human AS-IgG ELISA Kit from Assay Genie is an indirect immunoassay that uses biotin-streptavidin signal amplification to detect IgG-class anti-sperm antibodies in human specimens. Anti-sperm antibodies of the IgG isotype are clinically relevant markers investigated during the workup of immunological infertility, where circulating immunoglobulins can interfere with sperm motility, cervical mucus penetration, or fertilization.

Because the assay is indirect, microplate wells are pre-coated with purified sperm antigen preparations; serum samples or EDTA-plasma aliquots are added, and any anti-sperm IgG present binds to the immobilized antigens. A biotinylated anti-human IgG conjugate is then introduced, followed by streptavidin-HRP, allowing researchers to read the enzyme-substrate reaction with standard absorbance plate readers used in ELISA workflows.
