Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative detection of human amyloid-beta precursor protein (APP) in research samples. The assay uses a double-antibody sandwich format with colorimetric readout and is reactive with human samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. APP is a key protein in Alzheimer's disease pathogenesis, as it is cleaved to produce amyloid beta peptides that accumulate as plaques in the brain, making the kit a useful tool for studying neurodegenerative disorders and investigating potential treatments.

The reported sensitivity is 0.563 ng/mL with an assay range of 0.938-60 ng/mL. Reported linearity across 1:2, 1:4, and 1:8 dilutions falls within 80-105% for serum (85-104%, 86-99%, 89-105%), EDTA plasma (82-97%, 82-99%, 84-101%), and heparin plasma (80-99%, 87-100%, 83-100%). Recovery ranges from 87-101% (average 94%) in serum, 88-105% (average 97%) in EDTA plasma, and 97-103% (average 99%) in heparin plasma. Unopened kits are stored at 2-8°C for 12 months.

The protocol involves equilibrating reagents, loading standards and samples, incubating at 37°C for 90 minutes for primary antigen binding, adding biotin-labeled detection antibody with a 60-minute incubation, adding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after stop solution. Associated synonyms for the target include APP, ABPP, APPI, Alzheimer disease amyloid protein, Amyloid precursor protein, Amyloid-beta A4 protein, Cerebral vascular amyloid peptide, CVAP, PreA4, Protease nexin-II, PN-II, N-APP, Soluble APP-alpha, and S-APP-alpha (UniProt P05067).
