Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of human Apolipoprotein B (APOB) using a double-antibody format with colorimetric detection. The assay offers a sensitivity of 1.875 ng/mL across a working range of 3.125–200 ng/mL, and is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. Reagents should be stored at 2–8°C, where they remain stable for up to 12 months.

The procedure involves equilibrating reagents and the TMB substrate to room temperature, loading standards and samples onto the pre-coated plate, and incubating at 37°C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes. After TMB substrate development in the dark for 10–20 minutes, the reaction is stopped and absorbance is read at 450 nm. Guidance is provided for preparing serum, plasma, tissue homogenates, cell culture supernatant and cell lysates.

Apolipoprotein B is a key protein in lipid metabolism, responsible for transporting cholesterol and other fats in the bloodstream. Abnormal APOB levels have been associated with cardiovascular conditions including atherosclerosis and hyperlipidemia, making the kit a useful research tool for studies into lipid-related disorders.
