Human Apelin ELISA Kit

Manufactured by Assay Genie, this competitive ELISA is designed for the quantitative measurement of apelin (APLN) in human samples. The microtiter wells are pre-coated with apelin antigen, and the assay relies on a biotinylated detection antibody followed by HRP-Streptavidin and a TMB substrate; the colour intensity is inversely proportional to apelin concentration and absorbance is read at 450 nm using a standard microplate reader.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 37.5 pg/mL and a working range of 62.5-4000 pg/mL. Reported linearity values for serum (n=10) at 1:2, 1:4 and 1:8 dilutions are 87-102%, 86-102% and 94-105%; EDTA plasma ranges 82-98%, 85-94% and 82-100%; heparin plasma ranges 87-95%, 95-100% and 83-99%. Recovery averages are 96% for serum, 98% for EDTA plasma and 95% for heparin plasma.

Components are stored at 2-8°C and the kit is shipped on ice packs; kits are intended for research use only. The protocol involves adding 50 µL of standard or sample with 50 µL of biotin-labelled antibody, incubating at 37°C for 45 minutes, washing, developing with TMB substrate in the dark at 37°C for 10-20 minutes, stopping the reaction, and reading absorbance at 450 nm.
