Apurinic/apyrimidinic endonuclease 1 (APE1), the protein product of the APEX1 gene, is a central enzyme of the base excision repair pathway, responsible for processing abasic sites that arise from oxidative and alkylation DNA damage. Beyond its DNA repair activity, APE1 operates as a redox regulator of several transcription factors, including NF-κB, AP-1, and HIF-1α, linking genome maintenance to cellular signaling. This PharmaGenie ELISA kit from Assay Genie provides a quantitative immunoassay platform for measuring APE1 protein concentrations in human specimens.

Compatible with cell culture supernatant, plasma, and serum samples, the kit supports up to 96 individual assay determinations using a microplate-based workflow. Each run includes a standard curve generated from recombinant APE1 reference material, allowing interpolation of unknown sample concentrations. Standard incubation, wash, and detection steps are carried out on the pre-coated plate, and results are read on a conventional microplate reader.

APE1 has become a focus of translational research in oncology due to its overexpression in multiple tumor types and its association with chemoresistance. Quantification by ELISA complements downstream applications such as Western blot validation, immunohistochemistry of tumor sections, and functional DNA repair activity assays. The 96-assay format offers flexibility for cohort comparisons, dose-response studies, or time-course experiments examining APE1 induction following genotoxic stress in vitro.
