Description

A Sandwich ELISA (Double Antigen) from Assay Genie using a colorimetric detection method for the quantitative measurement of Anti-Survivin in human samples. Survivin is a key protein known for its role in regulating cell division and inhibiting apoptosis, making it a valuable biomarker in cancer research. The assay shows a sensitivity of 1.875 ng/mL and operates across a range of 3.125–200 ng/mL, with absorbance read at 450 nm after TMB substrate development. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents should be stored sealed at 2–8°C and should not be cryopreserved.

The protocol involves a 90-minute primary incubation at 37°C, a 60-minute biotin-labeled antigen binding step, a 30-minute HRP-streptavidin incubation, and a 10–20 minute color development in the dark before adding stop solution. In spiked-matrix linearity testing, serum (n=5) gave recoveries of 82–100% at 1:2, 81–100% at 1:4, and 90–98% at 1:8; EDTA plasma (n=5) gave 82–98%, 82–92%, and 85–105% across the same dilutions; and heparin plasma (n=5) gave 85–100%, 87–101%, and 82–99%. Spiked recovery ranges were 86–103% for serum, 85–100% for EDTA plasma, and 85–105% for heparin plasma, with corresponding averages of 96%, 93%, and 90%.
