Human Anti-Oxidized Low Density Lipoprotein/OLAb ELISA Kit

A colourimetric Sandwich ELISA (Double Antigen) kit from Assay Genie is intended for the quantitative measurement of human Anti-Oxidized Low Density Lipoprotein (OLAb) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is suited to research applications in cardiovascular disease, lipid metabolism and oxidative stress, where oxidized low-density lipoprotein is studied as a contributor to atherosclerosis. Reported sensitivity is 0.563 mIU/mL with a working range of 0.938-60 mIU/mL.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks into the pre-coated wells, and incubating at 37°C for 90 minutes to allow antigen binding. A biotin-labelled antigen working solution is then added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin conjugate (SABC) for 30 minutes at 37°C. TMB substrate is added and incubated in the dark for 10–20 minutes; after adding stop solution, absorbance is read at 450 nm.

Sample preparation guidance covers serum (allow blood to clot, then centrifuge at 1000 × g for 20 minutes), plasma (collect in anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate (homogenise with PBS or lysis buffer, then centrifuge), cell lysate (lyse on ice and centrifuge), and cell culture supernatant (centrifuge at 2500 rpm for 5 minutes). Kits should be stored sealed at 2-8°C and not cryopreserved.
