Description

A Sandwich ELISA kit from Assay Genie designed for the quantitative detection of human anti-apolipoprotein antibodies (AAHA) in research samples. The kit supports studies of lipid metabolism and immune-mediated cardiovascular and autoimmune disorders, including atherosclerosis.

The double-antigen sandwich assay uses colorimetric detection, with a sensitivity of 1.875 ng/mL and a working range of 3.125–200 ng/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored sealed at 2–8°C and not cryopreserved.

The procedure runs through six stages: equilibration of reagents and TMB substrate to room temperature and loading of standards, samples and blanks; primary incubation at 37°C for 90 minutes; binding of a biotin-labelled antigen working solution at 37°C for 60 minutes; incubation with HRP–streptavidin conjugate at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and a stop reaction with absorbance read at 450 nm. Dilution linearity has been demonstrated for serum (87–103% at 1:2, 82–99% at 1:4, 86–100% at 1:8), EDTA plasma (86–102%, 85–90%, 82–98%) and heparin plasma (88–101%, 90–100%, 82–98%), with mean recoveries of 95% (serum), 94% (EDTA plasma) and 96% (heparin plasma).
