The double-antigen sandwich assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with reactivity confirmed for human specimens.

The kit offers a reported sensitivity of 0.938 ng/mL and an assay range of 1.563–100 ng/mL, with a colorimetric readout measured at 450 nm following TMB substrate development. Linearity has been demonstrated in serum (86–105% at 1:2, 86–103% at 1:4, 94–103% at 1:8), EDTA plasma (83–97%, 87–100%, 88–98%), and heparin plasma (84–98%, 85–97%, 86–98%). Recovery ranges from 86–106% in serum, 86–104% in EDTA plasma, and 85–104% in heparin plasma, with averages of 96%, 94%, and 96% respectively.

Unopened kits should be stored sealed at 2–8°C and not cryopreserved. The protocol includes a 90-minute primary incubation at 37°C, followed by 60 minutes with biotin-labeled antigen and 30 minutes with HRP-streptavidin, then 10–20 minutes of TMB colour development in the dark before stop solution and absorbance reading. ANNA-1 antibodies target neuronal nuclear antigens and are associated with paraneoplastic neurological syndromes, encephalitis, and sensory neuropathy, making the kit relevant to research into autoimmune neurological disease.
