The assay is intended for research use only and has been validated for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Specificity is confirmed against both natural and recombinant human AMOTL1.

Manufacturer-reported performance characteristics include a detection range of 0.156–10 ng/mL and a sensitivity of 0.078 ng/mL. Intra-assay precision is 4.2% CV and inter-assay precision is 7.8% CV. Linearity in serum (n=5) at 1:2, 1:4, 1:8 and 1:16 dilutions is 89–97%, 109–119%, 94–103% and 103–113%. EDTA plasma (n=5) shows 104–104%, 96–106%, 107–116% and 80–90%, while heparin plasma (n=5) gives 99–107%, 84–95%, 104–113% and 83–96% across the same dilutions. Spike-and-recovery averages 99% in serum (range 93–105%) and 101% in plasma (range 95–107%).

AMOTL1 is a peripheral membrane component of tight junctions (UniProt Q8IY63) that inhibits the Wnt/β-catenin signaling pathway, probably by recruiting CTNNB1 to recycling endosomes and so preventing its translocation to the nucleus.
