The Phosphotyrosine CSK PharmaGenie ELISA Kit from Assay Genie provides a quantitative immunoassay for measuring tyrosine-phosphorylated CSK (C-terminal Src kinase), a non-receptor tyrosine kinase of approximately 50 kDa that negatively regulates Src-family kinase activity through phosphorylation of a conserved C-terminal tyrosine. Because phosphorylation of CSK is dynamically regulated by upstream signaling inputs, this phosphorylation ELISA is useful for studying immune-cell receptor signaling, integrin-mediated adhesion, and growth-factor responses where Src-family activity must be tightly controlled.

The kit is configured as a complete microplate-format ELISA with pre-coated wells, detection antibodies, and reagents sufficient for 96 assays. It is validated for use with cell lysates and tissue lysates derived from human and mouse samples, allowing direct comparison of CSK activation between species in co-culture or xenograft studies. Prepared lysates should be generated in lysis buffers supplemented with phosphatase inhibitors and sodium orthovanadate to preserve the phosphotyrosine epitope on the CSK protein that is recognized by the capture reagent.

Researchers in oncology, immunology, and cell-signaling labs can apply this ELISA as a higher-throughput complement to Western blot phospho-CSK blots, reading absorbance on a standard microplate reader. Each plate supports multiple standard-curve points and replicates, supporting quantitative interpolation of CSK phosphorylation levels across experimental conditions such as inhibitor titration, time-course stimulation, or comparison of wild-type and mutant backgrounds.
