Human and Mouse Phospho-SHP1 (S591) and Total SHP1 PharmaGenie ELISA Kit (SBRS1965)

Beyond pathway profiling, this kit is widely used as a phosphatase-activity surrogate in inhibitor screens against SHP1, a key negative regulator of JAK/STAT, B-cell receptor, and T-cell receptor signaling that is recurrently dysregulated in B-cell malignancies and insulin-resistant tissues. The dual-well configuration uses an anti-pan SHP1 capture antibody pre-coated on removable 8-well strips, so phospho-S591 and total SHP1 signals can be read from matched wells on the same plate.

Working range is 0.31 to 20 ng/mL with a 100 µL sample volume per well, and a treated Jurkat cell lysate is supplied as a positive control to verify capture and detection reagents in every run. The HRP/TMB reaction develops for 30 minutes at room temperature, with 50 µL of Stop Solution added before absorbance is measured on a standard microplate reader. For lysate preparation, cells are solubilized at 4 x 10^7 cells/mL in the supplied 2X Cell Lysate Buffer with protease and phosphatase inhibitors, incubated with shaking on ice for 30 minutes, and clarified at 13,000 rpm for 10 minutes before loading.
