The Assay Genie Phospho-PLCG2 (Tyr753) and Total PLCG2 PharmaGenie ELISA Kit enables simultaneous quantification of phosphorylated PLCG2 at tyrosine 753 and total PLCG2 protein from the same lysate, supporting pathway analysis in B-cell receptor, Fc receptor, and downstream immune signaling studies. Phospholipase C Gamma 2 is a key signaling effector in hematopoietic cells, and phosphorylation at tyrosine 753 reflects activation downstream of Syk and BTK in the receptor cascade.

This phosphorylation ELISA is supplied as a complete microplate kit configured for 96 assays per plate, with established use on human and mouse cell lysates and tissue lysates. Standard protocol typically uses approximately 100 µL of prepared sample per well, with bound analyte quantified using a chromogenic substrate incubation read on a standard microplate spectrophotometer. Component vials should be stored refrigerated on arrival; reconstituted standards and concentrated detection antibody should be aliquoted and frozen after first use, with unused strip wells resealed in the foil pouch with desiccant.

The dual-analyte detection format lets researchers normalize phospho-PLCG2 signals against total PLCG2, controlling for sample-to-sample variation in cell number, lysis efficiency, or baseline expression. This normalization is particularly useful when comparing activation states across immune cell populations, screening kinase inhibitors or small-molecule modulators, and probing downstream pathway activity in autoimmune disease models and related in vitro research.
