Human Amyloid Beta 40/AB 1-40 ELISA Kit

Assay Genie supplies a colorimetric Sandwich ELISA for the quantitative detection of human amyloid beta 40 (AB 1-40), a protein associated with plaque formation in Alzheimer's disease and other neurodegenerative disorders. The assay uses a double-antibody sandwich configuration and is reactive with human samples.

Performance parameters include a sensitivity of 4.688 pg/mL and an assay range of 7.813-500 pg/mL. Compatible sample types are serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2-8°C for 12 months.

Linearity and recovery were evaluated in serum, EDTA plasma, and heparin plasma (n = 5 each). Linearity for serum spans 85-104% at 1:2, 84-97% at 1:4, and 82-98% at 1:8 dilution; EDTA plasma shows 85-105%, 84-97%, and 86-100% across the same dilutions; heparin plasma shows 88-100%, 82-98%, and 82-94%. Recovery ranges from 86-100% (average 95%) in serum, 88-105% (average 95%) in EDTA plasma, and 87-105% (average 96%) in heparin plasma. The workflow uses plate and reagent equilibration, a 90-minute primary incubation at 37°C, biotin-labelled detection antibody binding for 60 minutes, HRP-Streptavidin incubation for 30 minutes, TMB color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution addition.
