The Assay Genie human AMY2 sandwich ELISA enables laboratories to quantify pancreatic alpha-amylase with sensitivity capable of detecting low-abundance enzyme levels in clinical research samples. AMY2 is the pancreatic isoform of alpha-amylase (EC 3.2.1.1), a calcium-dependent glycoside hydrolase that cleaves internal alpha-1,4 linkages in starch and glycogen to release maltose and oligosaccharides. Because salivary (AMY1) and pancreatic (AMY2) isoforms differ by only a few amino acids, isoform-specific immunoassays like this kit are required when researchers need to distinguish pancreatic output from total circulating amylase activity.

Typical research applications include studies of acute pancreatitis, exocrine pancreatic insufficiency, cystic fibrosis-related pancreatic dysfunction, and metabolic investigations linking pancreatic enzyme secretion to glycemic control and incretin responses. The pre-coated 96-well microplate format supports up to 96 assays per kit, with sample volumes usually loaded in the microliter range and standards run in duplicate to construct a multi-point standard curve, commonly spanning roughly 0.1-10 ng/mL depending on the recombinant calibrator dilution series.

For best results, store unopened reagents at 2-8 °C and keep the lyophilized standard frozen at -20 °C in single-use aliquots to limit repeated freeze-thaw cycles, as the recombinant calibrator is the most labile component. Bring all reagents to room temperature before use and protect the streptavidin-HRP conjugate and TMB substrate from light. Researchers should treat all human-derived samples as potentially biohazardous and perform the assay in vitro under standard BSL-2 practices, with results interpreted in the context of paired clinical data.
