Description

Designed for the precise measurement of amphiphysin levels in human samples, this Sandwich ELISA kit from Assay Genie uses a Double Antigen format with colorimetric detection. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay offers a sensitivity of 18.75 pg/mL across a working range of 31.25-2000 pg/mL.

Amphiphysin is a key protein involved in synaptic vesicle endocytosis, playing a critical role in neurotransmission. Dysregulation has been linked to neurological disorders such as Alzheimer's disease and multiple sclerosis, making this anti-AMPH kit a relevant tool for cell biology and neurobiology research.

Linearity at 1:2, 1:4 and 1:8 dilutions was 85-94% / 83-93% / 84-99% for serum, 85-95% / 84-101% / 82-99% for EDTA plasma, and 91-105% / 90-101% / 84-99% for heparin plasma (n=5). Recovery ranges were 85-104% (average 94%) for serum, 88-103% (average 93%) for EDTA plasma, and 88-105% (average 95%) for heparin plasma. The workflow involves equilibrating reagents and TMB substrate, loading standards, samples and controls on the pre-coated plate and incubating at 37°C for 90 minutes, adding biotin-labeled antigen working solution for 60 minutes, then HRP-Streptavidin for 30 minutes, developing colour with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution. Kits should be stored sealed at 2-8°C and not cryopreserved.
