Vascular adhesion protein-1 (VAP-1), also called semicarbazide-sensitive amine oxidase (SSAO) and encoded by the AOC3 gene, is a copper-dependent amine oxidase anchored to vascular endothelium that supports leukocyte rolling and deaminates primary amines. The Assay Genie human AOC3 ELISA kit provides laboratories with a quantitative tool to measure this shed, circulating form of the enzyme across research samples and experimental models.

The assay is calibrated against recombinant human AOC3, supporting inter-laboratory reproducibility when standard curves are freshly prepared. Each kit is supplied with pre-coated microplate wells, biotinylated detection antibody, recombinant standards, streptavidin-HRP reagents, substrate solution, and wash and assay buffers in a configuration sufficient for 96 determinations.

Investigators working on chronic inflammatory disorders, vascular dysfunction, type 2 diabetes complications, hepatic steatosis, and selected solid-tumor microenvironments use circulating AOC3 as a candidate biomarker. The protocol accommodates human serum, EDTA-plasma preparations, and clarified tissue-homogenate inputs, making the kit versatile for patient-cohort studies, cell-based experiments on cultured endothelial monolayers stimulated with inflammatory cytokines, and expression profiling in lysates from inflamed versus control tissues.

Representative applications include measuring SSAO activity surrogates in longitudinal patient-cohort sera, tracking VAP-1 release from activated endothelium under TNF-alpha or IL-1beta stimulation, and comparing AOC3 levels across biopsies from inflammatory versus non-inflammatory tissue sites.
