Description

A sandwich ELISA, double antibody kit from Assay Genie for the quantitative measurement of human AMD1 (S-adenosylmethionine decarboxylase proenzyme). The assay uses a pre-coated capture antibody on a 96-well plate, a biotin-conjugated detection antibody, and HRP-Streptavidin with TMB substrate, with absorbance read at 450 nm. Reactivity is human, and the kit is intended for research use only.

According to the manufacturer datasheet, the assay has a reported sensitivity of 0.375 ng/mL and a range of 0.625–40 ng/mL. Reported linearity for serum (n=5) is 86–98% at 1:2, 90–103% at 1:4 and 87–92% at 1:8; for EDTA plasma (n=5) 88–91%, 85–98% and 83–90%; and for heparin plasma (n=5) 82–99%, 82–91% and 82–100%. Reported recovery averages are 93% for serum, 96% for EDTA plasma and 92% for heparin plasma. The kit is shipped on ice packs and reagents are stored at 2–8°C.

Compatible sample types listed by the manufacturer include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sample preparation guidance specifies centrifugation of serum at 1000 × g for 20 minutes, plasma collection with anticoagulant and centrifugation at 1000 × g for 15 minutes at 2–8°C, and tissue homogenisation in PBS with protease inhibitors. AMD1 is described in the product literature as a key enzyme in spermidine and spermine biosynthesis, with dysregulation linked in the source text to cancer, metabolic disorders and inflammatory conditions.
