Description

This sandwich ELISA kit from Assay Genie is designed for the quantitative detection of Alpha-methylacyl-CoA racemase (AMACR) in human biological samples. AMACR is an enzyme involved in fatty acid metabolism and is often overexpressed in prostate cancer and other malignancies, so precise measurement of its levels can support research into disease progression and potential treatment options.

It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity in serum falls between 88–102%, 87–104% and 87–104% at 1:2, 1:4 and 1:8 dilutions respectively, while EDTA plasma yields 82–96%, 82–91% and 87–99%, and heparin plasma yields 87–102%, 82–94% and 81–99%. Recovery ranges are 86–102% for serum (average 92%), 86–103% for EDTA plasma (average 94%) and 91–103% for heparin plasma (average 95%). All reagents are stored at 2–8°C for 12 months.

The protocol begins with reagent equilibration to room temperature, followed by loading standards, samples and a zero control onto the pre-coated plate. The plate is incubated at 37°C for 90 minutes for antigen binding, then sequentially with a biotin-labelled detection antibody (37°C, 60 minutes) and HRP-streptavidin conjugate (37°C, 30 minutes). TMB substrate is added and developed in the dark for 10–20 minutes before stop solution is introduced and absorbance is read at 450 nm. Supplied components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-streptavidin conjugate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, wash buffer, stop solution and plate sealers.
