Description

Assay Genie provides a sandwich ELISA, double-antibody kit for the quantitative measurement of human bone alkaline phosphatase. The colorimetric assay is intended for research use with plasma, cell culture supernatant, cell or tissue lysate and other liquid samples, and the manufacturer's description additionally references serum. It targets ALPL (bone alkaline phosphatase), an enzyme involved in bone mineralization and turnover and a biomarker of interest in conditions such as osteoporosis and Paget's disease.

Reported analytical performance includes a sensitivity of 1.875 ng/mL and a standard range of 3.125–200 ng/mL. In dilution linearity testing, EDTA plasma (n = 5) gave recoveries of 85–101% at 1:2, 83–96% at 1:4 and 83–95% at 1:8, while heparin plasma (n = 5) gave 85–99% at 1:2, 95–101% at 1:4 and 81–89% at 1:8. Spike-and-recovery experiments reported average values of 95% for EDTA plasma (range 89–104%, n = 5) and 95% for heparin plasma (range 85–102%, n = 5). The workflow uses a pre-coated microplate: standards and samples are incubated at 37 °C for 90 minutes, followed by a biotinylated detection antibody for 60 minutes at 37 °C, HRP-streptavidin for 30 minutes at 37 °C, and TMB substrate for 10–20 minutes in the dark. The reaction is stopped and absorbance read at 450 nm. Reagents are stored at 2–8 °C for 12 months. The kit is associated with UniProt entry P05186 and the synonyms BALP, Bone Alkaline Phosphatase, ALPL, BSAP, HOPS, AP-TNAP and TNSALP.
