Beyond serum and plasma, this ELISA has been applied to cerebrospinal fluid (CSF) and conditioned cell culture medium in published studies, the former being especially relevant for Alzheimer's disease and other neurodegenerative research where SERPINA3 expression in the central nervous system is of interest. A standard 96-well plate layout accommodates up to 40 unknowns in duplicate alongside the standard curve, and most users complete the workflow in roughly 2.5–3 hours at room temperature once samples are prepared.

For sample preparation, serum and plasma are typically tested neat or at a 1:2 dilution in the supplied sample dilution buffer, while CSF and conditioned medium often require further dilution to fall within the 1.5–100 ng/mL working range. The minimum detectable dose is generally reported near 0.5 ng/mL, giving a dynamic range spanning roughly two orders of magnitude. Researchers profiling acute-phase responses often run this kit alongside CRP or SAA assays to characterize systemic inflammation.

Each unopened kit from Assay Genie remains stable for approximately 6 months when kept refrigerated at 2–8 °C; once the lyophilized standards and streptavidin-HRP working solution are reconstituted, aliquot and freeze them at -20 °C to limit freeze-thaw cycles, and protect the HRP conjugate from light. Bring all reagents to room temperature (approximately 20–25 °C) before pipetting to prevent condensation in the wells and to maintain consistent kinetics during the TMB incubation step.

The strip-well format is compatible with any standard microplate reader equipped with a 450 nm primary filter and an optional 570 or 630 nm reference filter for background subtraction, and it allows partial-plate runs for lower-throughput experiments while still supporting the full 96 determinations when needed.
