Description

Using a double-antibody sandwich ELISA format, the Assay Genie kit provides colorimetric detection of alpha-lactalbumin in human biological samples with a reported sensitivity of 0.375 ng/mL and a measuring range of 0.625-40 ng/mL. Alpha-lactalbumin is a key protein found in human milk that plays a critical role in infant nutrition and development, and has been investigated in research contexts relating to cancer, immune disorders, and neurological diseases.

Acceptable sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and the kit is stored at 2-8°C for 12 months. Linearity for serum (n=5) is 98-105% at 1:2, 89-100% at 1:4, and 87-105% at 1:8; EDTA plasma 82-99%, 82-96%, and 88-100%; and heparin plasma 85-98%, 81-94%, and 89-100%. Recovery ranges averaged 92% for serum, 98% for EDTA plasma, and 93% for heparin plasma.

The workflow involves equilibrating reagents, loading standards and samples for primary incubation at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes at 37°C, binding HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing colour with TMB substrate in the dark for 10–20 minutes, then adding stop solution and reading absorbance at 450 nm.
