Human alpha 2 Macroglobulin ELISA Kit

Manufactured by Assay Genie, this double-antibody sandwich ELISA is designed for the quantitative measurement of human alpha-2 macroglobulin using a colorimetric detection method. The assay is reactive with human samples and has been validated for use with serum, plasma, cell culture supernatant, and cell or tissue lysate.

Performance specifications include a sensitivity of 46.875 ng/mL and a standard curve range of 78.125–5000 ng/mL. All reagents are stored at 2–8°C and remain stable for 12 months. The workflow involves equilibrating reagents and the TMB substrate to room temperature, loading standards and samples onto the pre-coated plate for a 90-minute incubation at 37°C, adding the biotin-labelled detection antibody for 60 minutes, applying HRP-Streptavidin for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, then stopping the reaction and reading absorbance at 450 nm.

Linearity has been verified in serum, EDTA plasma, and heparin plasma across 1:2, 1:4, and 1:8 dilations, with recovery averaging 97–98% across all three matrices.
